Notch pathway structure

Crystal structure of activated Notch, CSL and MAML

The Notch signaling pathway and MAML co-activators play central roles in cancer development and progression. Notch signaling is highly context-dependent and regulates critical cellular fate decisions, proliferation, and differentiation. Dysregulation of this pathway contributes to oncogenesis across multiple malignancies.

Ligand binding triggers a series of proteolytic cleavages, NICD nuclear translocation, and gene activation. Multiple nodes in this pathway are targetable, including ligand interactions, NRR stabilization, post-translational modifications, cleavage events, trafficking mechanisms, and protein-protein interactions.

MAML proteins scaffold the NICD–CSL complex and recruit co-activators; without MAML, transcription terminates. Notch hyperactivation can drive proliferation, impaired differentiation, apoptosis resistance, tumor microenvironment interactions, and cancer stem cell maintenance. In some tissue contexts, Notch may be tumor-suppressive, suggesting potential reactivation strategies.
